Clinical Evaluation of PCR for Rapid Detection of Methicillin-resistant Staphylococcus aureus


Abstract
In order to improve the detection rate and rapidity in detection of the mec A gene of methicillinresistant S. aureus(MRSA), we evaluated the sensitivity and reproducibility of detection with varied cell density and DNA preparation methods using
clinical
sources. The entire reaction time took about 5 hours, enabling this method to be used as a routine method in a clinical laboratory. Among three DNA preparation methods compared, lysostaphin method was most stable and the detection of the mec A
gene
was
possible when bacterial cells were present at a concentration of 10¡¿10E8 CFU/ml and above in the case of MRSA. In the clinical isolates of S. aureus, all 40 strains of MRSA were positive for mec A gene, whereas all 40 strains of methicillin
sensitive
S. aureus(MSSA) were negative. These results suggest that the modified PCR method is suitable for a routine clinical laboratory, and proper bacterial suspension and DNA preparation with lysostaphin are recommended to be used in direct detection
of
the
mec A gene.
